Semaglutide (Wegovy)
- Titration
- Weekly subcutaneous injection, starting at 0.25 mg. Dose is increased roughly every 4 weeks — 0.25 mg → 0.5 mg → 1 mg → 1.7 mg → 2.4 mg — over about 16 weeks, to reduce GI side effects.
- Maintenance dose
- 2.4 mg once weekly (1.7 mg is an FDA-approved alternative maintenance dose for patients who don't tolerate 2.4 mg).
- Trial results
- In the STEP 1 trial (68 weeks), participants on semaglutide plus lifestyle intervention lost a mean of 14.9% of body weight, vs. 2.4% on placebo.
